丹参酮ⅡA对2型糖尿病患者外周血中SDF-1/CXCR4的影响

摘要:

目的探讨丹参酮ⅡA(TanⅡA)对2型糖尿病(T2DM)患者外周血中基质细胞衍生因子-1(SDF-1)和其受体CXCR4的影响。方法62例T2DM患者随机分为对照组和实验组。对照组采用常规治疗;实验组在常规治疗基础上加用TanⅡA40mg.d-1,持续2周。测定治疗前后所有对象的外周血中SDF-1浓度、内皮祖细胞(EPCs)数量及其表面CXCR4阳性率。结果治疗前,两组各项参数比较差异无统计学意义(P>0.05)。治疗前后比较,对照组各参数差异无统学意义(P>0.05)。实验组治疗后较治疗前SDF-1、CXCR4、EPCs上升,差异有统计学意义(P<0.05或P<0.01);且EPCs与SDF-1呈显著性正相关(r=0.223,P<0.05)。结论TanⅡA可升高T2DM患者外周血中SDF-1水平和上调CXCR4的表达,从而增加EPCs数量,改善内皮功能。

Abstract:

AIM To study effect of TanshinoneⅡA(TanⅡA) on stromal cell-derived factor-1(SDF-1) and it's receptors CXCR4 in patients with type 2 diabetes mellitus(T2DM).METHODS Sixty-two patients with T2DM were randomly divided into treatment and control groups.TanⅡA 40 mg·d-1 was given to patients in treatment group for 2 weeks.The level of plasma SDF-1,the positive rate of CXCR4 on endothelial progenitor cells(EPCs)and the number of EPCs were determined pre-and post-treatment.RESULTS All parameters had no significant differences between 2 groups before observation(P>0.05).Increasing SDF-1,EPCs and CXCR4 had significant difference in the treatment group pre-and post-treatment(P<0.05 or P<0.01).The parameters had no significant differences between before and after observation in the control group(P>0.05).There was a significant positive correlation between EPCs and SDF-1(r=0.223,P<0.05).CONCLUSION TanⅡA can improve vascular endothelial function in T2DM,which may be due to elevating the level of plasma SDF-1 and increasing the expression of CXCR4 on EPCs,and inducing EPCs mobilization from bone marrow to peripheral blood.
