HPLC法测定翁沥通胶囊中木犀草苷、芦荟大黄素、栀子苷和甘草苷的含量

摘要:

目的 建立HPLC法用于测定翁沥通胶囊药材组成中木犀草苷、芦荟大黄素、栀子苷和甘草苷4种成分的含量。方法 采用Agilent ZORBAX Eclipse Plus C18色谱柱分离,柱温为室温,流速设定为1.0 mL·min-1,检测波长:木犀草苷351 nm、芦荟大黄素254 nm、栀子苷和甘草苷237 nm,进样量为10μL,采用HPLC行定性、定量检测。结果 木犀草苷、芦荟大黄素、栀子苷和甘草苷分别在质量浓度为0.039 24~3.924 00μg·mL-1、0.039 00~3.900 00μg·mL-1、0.029 43~2.943 00μg·mL-1和0.019 04~1.904 00μg·mL-1内具有良好的线性;加标平均回收率分别为99.63%、100.41%、100.04%和99.84%;方法重复性RSD分别为0.98%、1.30%、1.00%和1.10%;供试品溶液在24 h内稳定。结论 实验建立的HPLC法具有前处理简单、分析时间短、检测结果准确等优点,适用于翁沥通胶囊的质量控制。

Abstract:

AIM A rapid high-performance liquid chromatography(HPLC) method was developed to detect the content of 4 components, namely luteoloside, aloe-emodin, geniposide and liquiritin, in the medicinal composition of Wenglitong capsules. METHODS Separation was performed using an Agilent ZORBAX Eclipse Plus C18 column maintained at room temperature, with a mobile phase flow rate of 1.0 mL·min-1. Detection wavelengths were set at 351 nm for luteoloside, 254 nm for aloe-emodin, and 237 nm for simultaneous monitoring of geniposide and liquiritin. An injection volume of 10 μL was employed, and qualitative and quantitative analyses were performed using HPLC detection. RESULTS Luteoloside, aloe-emodin, gardenin and liquiritin exhibited good linearity within the ranges of 0.0392 40-3.924 00 μg·mL-1, 0.0390 0-3.900 00 μg·mL-1, 0.0294 3-2.943 00 μg·mL-1 and 0.019 04-1.904 00 μg·mL-1, respectively. The average recovery rates were 99.63%, 100.41%, 100.04% and 99.84%, respectively. The reproducibility RSDs were 0.98%, 1.30%, 1.00% and 1.10%, respectively. The test solution remained stable for 24 h. CONCLUSION The findings indicate that the method offers advantages such as simple pretreatment, reduced analysis time, and precise detection results, which is suitable for the quality control of Wenglitong capsules.
