抗流感病毒药物联合应用的研究进展

摘要:

流行性感冒(简称“流感”)是由流感病毒引起的一种急性呼吸道传染病,传染性强,发病率高,具有全球分布特性,是受到全球关注的重要公共卫生问题。尽管流感多可自愈,但在高危人群中也可能引起严重并发症,甚至危及生命。在目前已知的4种类型的流感病毒中,甲型和乙型流感病毒在人群中呈季节性流行,其中甲型流感病毒危害性最大,其在全球范围内的检测率超过了乙型流感病毒。迄今为止,已有多种抗流感病毒药物应用于临床流感的治疗,但由于流感病毒的突变性和耐药性,现有抗流感病毒药物的有效性降低。抗流感病毒药物联合治疗具有潜在价值,不同作用机制的药物联合使用可能会增加药物的协同性,降低药物的耐药性,为临床提供新的药物治疗方案。该综述系统梳理了当前抗流感病毒药物的联用情况,探讨了药物联合应用的价值,为临床提供参考。

Abstract:

Influenza is an acute respiratory infectious disease caused by influenza viruses, characterized by high infectivity, high incidence, and global distribution. It is a major public health issue of global concern. Although influenza is often self-limiting, it may cause severe complications and even be life-threatening in high-risk populations. Among the 4 known types of influenza viruses, influenza A and B viruses are responsible for seasonal epidemics in humans, with influenza A virus posing the greatest threat. Its surveillance detection rate on a global scale has surpassed that of influenza B virus. Although multiple classes of antiviral agents against influenza have been deployed clinically, the efficacy of existing therapeutics is progressively compromised by viral mutation and the emergence of drug resistance. Combination therapy with antiviral agents against influenza represents a promising strategy, where the deployment of mechanistically distinct drugs may enhance synergistic effects, delay the emergence of resistance, and provide novel therapeutic approaches for advanced patient care. This review critically examines current combinatorial regimens of antiviral agents against influenza, explores the therapeutic value of drug synergy, and provides a reference for clinical practice.
