微波辅助萃取-高效液相色谱法测定金银花药材中槲皮素和木樨草素的含量

摘要:

目的建立微波辅助萃取-高效液相色谱(MAE-HPLC)法分析金银花药材中槲皮素和木樨草素的含量。方法采用Diamonsil C18柱(150 mm×4.6 mm,5μm),以甲醇-0.2%三氟乙酸(55∶45,V/V)为流动相,流速1.0 mL.min-1,检测波长350 nm。微波辅助萃取条件:2.5 mol.L-1盐酸甲醇溶液10 mL,微波功率为360 W,提取时间为3 min。结果槲皮素在5~100 mg.L-1内线性关系良好(r=0.999 5),平均加样回收率为92.35%,RSD为1.33%(n=3);样品的日内和日间RSD分别为0.44%和1.61%。木樨草素在0.255 mg.L-1内线性关系良好(r=0.999 9),平均加样回收率为92.40%,RSD为1.13%;日内和日间RSD为0.79%和2.60%。结论该方法快速、准确,操作便捷,适用于提取分析和测定金银花中槲皮素和木樨草素的含量。

Abstract:

AIM To develop a microwave assisted extraction-HPLC method for the determination of two flavonoids compunds of Flos lonicerae.METHODS Quercetin and luteolin in Flos lonicerae were determined by HPLC on Diamonsil C18 column(150 mm×4.6 mm,5 μm)with methanol-0.2% trifluoroacetic acid(55∶45,V/V)as the mobile phase.The detection wavelength was 350 nm.The flow rate was 1.0 mL·min-1.Quercetin and luteolin were both extracted by microwave assisted extraction with 10 mL 2.5 mol·L-1methanol-chlorhydric acid using condition of 360 W and 3 min.RESULTS The linear range of quercetin was 0.25-5 mg·L-1(r=0.999 9) and the linear range of luteolin was 5-100 mg·L-1(r=0.999 5).The average recovery and RSD value of quercetin were 92.35% and 1.33%(n=3);the average recovery and RSD value of luteolin were 92.40% and 1.13%(n=3),respectively.The RSD of intra-day and inter-day of quercetin were 0.44% and 1.61%,respectively;the RSD of intra-day and interday of luteolin were 0.79% and 2.60%,respectively.CONCLUSION The method is simple,precise and accurate,and can be used to determine and analyze quercetin and luteolin in Flos lonicerae.
