目的观察金黄色葡萄球菌(金葡萄)滤液制剂对机体免疫功能的影响。方法采用小鼠单次腹腔注射环磷酰胺60mg·kg-1造成免疫功能低下动物模型,以2、4和6μg·kg-1金葡菌滤液制剂为受试药物,药物剂量以检品中含有的金葡菌肠毒素C表示。采用经口灌胃给药途径,连续给药10d,停药后24h处死动物并进行各项免疫指标检测。结果小鼠腹腔注射环磷酰胺60mg·kg-1可明显降低实验小鼠的细胞和体液免疫功能,本品可明显提高免疫功能低下模型小鼠的免疫功能。其中包括脾重量指数、半数溶血值(HC50)、ConA诱导的脾淋巴细胞转化能力、NK细胞活性、血清白细胞介素Ⅱ(IL-2)以及血清肿瘤坏死因子(TNF)等项测定,给药各剂量组均明显高于模型对照组,统计学比较差异有显著或非常显著意义。结论金葡菌滤液制剂经口给药,可使因注射环磷酰胺所致的免疫功能低下模型小鼠的细胞免疫和体液免疫功能得到很好的改善和提高,其中以细胞免疫功能的改善和提高更为明显。
Abstract:AIM To observe the effect of the filter preparation of Staphylococcus aureus (FPSA)on organism immunity function. METHODS The mice were intraperitoneally injected with cyclophosphamide in single dose of 60 mg·kg -1 to create the low immunity function animal model. The FPSA in dose of 2, 4 and 6 μg·kg -1 were took as the agents, and the dosage expressed the content of staphylococcal enterotoxinic(SEC). The mice were administered the agents orally for 10 d, then several immunity indexes were determined after 24 h recovery period. RESULTS The FPSA approved to obviously improve the low immunity function induced by cyclophosphamide in dose of 60 mg·kg -1.The spleen ponderal index, 50% serum hemolysis (HC_ 50), spleen lymphocyte transformation ability induced by ConA , NK cell activeness, the interleukin-Ⅱ(IL-2),tumor necrosis factor(TNF) were measured. Significant differences were noted between the control group and 3 dose groups. The indexes in 3 dose groups were all obviously higher than those in the control group. CONCLUSION The FPSA approved to obviously improve the low immunity function induced by cyclophosphamide, especially the cellular immunity function.
