目的采用一个简单的、实用的方法评价临床个体患儿体内CYP3A酶活性;预测儿童白血病患者体内依托泊苷血药浓度、药动学参数。方法采用HPLC测定药物浓度,一点法计算药物消除半衰期,尿样法测定体内CYP3A酶活性。结果儿童白血病患儿体内依托泊苷药物浓度与CYP3A酶活性相关性较差,而其t1/2与个体在化疗前CYP3A酶活性变化的相关性较好。结论依托泊苷t1/2与儿童白血病患儿体内CYP3A酶活性有一定的相关性。
Abstract：AIM To establish a simple and practical method for the estimation of the inter-patient variability of cytochrome P450(CYP3A)enzyme activity and to predict drug concentration and parmacokinetics of etoposide with the variability of CYP3A in chidren patients with leukemia.METHODS The drug concentration of etoposide was determined by HPLC, drug eliminated half life was calculated by one point method and CYP3A enzyme activity was determined by urine sample method.RESULTS Relativity analysis between drug concentration of etoposide and CYP3A enzyme activity in patients with leukemia was poor correlated,but it was having a good correlation between drug eliminated half life of etoposide and the variability of CYP3A enzyme activity.CONCLUSION Drug eliminated half life of etoposide and the variability of CYP3A enzyme activity have a good correlation.
