目的 :探讨金蒲抑瘤片体外对肝癌细胞的诱导凋亡作用。方法 :应用MTT法、生长曲线、荧光染色、血清药理学、DNA凝胶电泳等实验技术研究金蒲抑瘤片体外对BEL 74 0 4细胞的增殖抑制和诱导细胞凋亡的作用。结果 :金蒲抑瘤片体外对BEL 74 0 4细胞有明显的抑制作用 ,且这种抑制作用呈浓度依赖性。经体内代谢的含药血清也具有抑制细胞生长的能力。药物作用后的细胞出现荧光染色增强、胞核碎裂、凋亡小体形成等凋亡形态学变化。结论 :金蒲抑瘤片及其含药血清 ,在一定浓度下能诱导BEL 74 0 4细胞凋亡

AIM: To investigate the effects of Jinpu inhibitive tumor tablets on induction of apoptosis in human liver cancer cell line BEL 7404. METHODS: MTT colorimetric assay was used to examine the growth inhibition of BEL 7404 cells. Cell morphology and DNA gel electrophoresis were used to observe the apoptosis. RESULTS: The solution of Jinpu inhibitive tumor tablets showed dose dependent inhibitory effects on the proliferation on BEL 7404 cell line. Cells treated with 0 5 mg·mL -1 and 0 2 mg·mL -1 Jinpu tablets for 24 h displayed DNA ladder bands and typical morphological change of apoptosis. Jinpu inhibitive tumor tablets serum can inhibit the BEL 7404 cells prolieration, too. CONCLUSION: Jinpu inhibitive tumor tablet has growth inhibiting effect and may induce apoptosis in BEL 7404 in a certain dose range.
