毛细管气相色谱法测定大鼠血浆中β-苯基乳酸浓度

摘要:

本文报道采用SE-54石英毛细管色谱柱及电子捕获检测器(63Ni-ECD)毛细管气相色谱法(CGC)测定了大鼠血浆中β-苯基乳酸(PLA)的浓度,线性范围1.6~9.6ng,相关系数r=0.9997,检测限为0.5ng;采血量为0.1ml,并用醋酸乙酯对大鼠血浆中PLA进行提取,方法灵敏,回收率高,平均回收率为99.04%。日内与日间平均变异系CV分别为5.1272%,5.9974%。此方法可用于临床药代动力学研究。

Abstract:

The determination of β-Phenylactic acid (PLA) in rat's plasma by capillary chromatography is described. Quantitation was effected by 63 Ni-electron capture detector after separation with a SE-54 quartz capillary chromatographic column. Claibration curves of the instrumental responses against drug concentrations were linear within the range 1. 6-9. 6ng. The limit of detection of PLA wae considered to be 0. 5ng. PLA was extracted from plasma with ethyl acetate (rat's plasma 0.1ml). The recovery from plasma was higher and the mean valune was 99. 04%. Their coefficents of variation (CV) of within day and between day were 5. 1272%, and 5. 9974% respectively.
