摘要:

目的采用HPLC法对六神丸中蟾酥的主要成分华蟾酥毒基和酯蟾毒配基进行含量测定。方法样品经甲醇回流提取,采用HPLC法测定。色谱柱为Hypersil C18(250 mm×4.6 mm,5μm);流动相为乙腈-水(43:57,V/V);流速1.0 mL·min-1;检测波长296 nm;温度25℃。结果华蟾酥毒基为0.052 5~1.050μg(r=0.999)、酯蟾毒配基为0.052~1.040μg(r=0.999)时呈良好的线性关系。华蟾酥毒基平均加样回收率为100.18%,RSD=1.85(n=6);酯蟾毒配基平均加样回收率为100.40%, RSD=1.14(n=6)。结论此方法简单、快速、准确,可用于六神丸中华蟾酥毒基和酯蟾毒配基含量测定。

Abstract:

AIM To determine quantitatively cinobufagin and resibufogenin for chief component of venenum bufo-nis in liushen pill by HPLC. METHODS The column was Hypersil C18 (250 mm×4.6 mm, 5μm); the mobile phase was acetonitrile-water (43:57, V/V) ; the flow rate was 1.0 mL·min-1; the detective wavelength was 296 nm; the column temperature was 25℃. RESULTS The linear range of cinobufagin was 0.052 5 - 1.050μg( r = 0.999) , the average recovery was 100.18% ,and RSD - 1,85( n = 6) ;the linear range of resibufogenin was 0.052 - 1.040μg( r = 0.999), the average recovery was 100.40% ,and RSD = 1.14( n = 6). CONCLUSION The method is simple, rapid, accurate, and can be applied to the determination of cinobufagin and resibufogenin in liushen pill.
