目的研究不同溶剂及浓度对血竭体外透皮吸收的影响,优化血竭经浅表创伤皮肤的给药方案。方法采用改良的Franz扩散池装置,以针刺模拟浅表创伤大鼠皮肤为屏障,运用HPLC法测定接收液中的血竭素含量,计算累积透过率和透皮吸收速率常数(J),确定血竭的最佳给药剂型和剂量。结果以75%乙醇溶液为最佳溶剂,以5%氮酮作为促透剂,血竭最佳质量浓度为27.5g·L-1,最佳剂量为82.7mg·cm-2;优化制剂在8h内稳定渗透释放,J为0.027mg·cm-2·h-1。结论血竭75%乙醇溶液可作为临床伤口护理的最佳制剂。
Abstract:AIM To investigate the effect of different dosage forms and concentrations on percutaneous absorption of Draconis Sanguis in vitro,and optimize the dosage regimens of skin with superficial wound.METHODS Modified Franz diffusion cell device was chosen,and needled skin of SD rat which was simulated superficial wound was used as cutaneous barrie.The concentration of dracorhodin in receiving solution was determined by HPLC,and the optimal dosage form and dose were confirmed by calculating the accumulated transdermal rate and the rate constant of percutaneous absorption.RESULTS With the 75% ethanol solution as the solvent,and 5% azone solution as penetration enhancer,the best concentration of Draconis Sanguis was 27.5g·L-1,and the best dose was 82.7mg·cm-2.The optimized formulation was released stably in 8h,and the rate constant of percutaneous absorption J was 0.027mg·cm-2·h-1.CONCLUSION The dosage form of 75%ethanol solution of DraconisSanguisis superior to the other dosage forms,and it can be used as the treatment of superficial wound in clinic.
